Il destino della galassia è nelle mani di due accaniti rivali, provenienti da mondi assai diversi fra loro. Da un lato, James Tiberius Kirk (Chris Pine), un ribelle ragazzo dell'Iowa assetato di emozioni, un leader nato in cerca di una causa da difendere. Dall'altro, Spock (Zachary Quinto), originario del pianeta Vulcano in cui domina la ragione pura, ma che, a causa della sua duplice natura umana e aliena, non è insensibile nei confronti di quelle emozioni che i vulcaniani hanno da tempo cancellato dalla loro vita. Spock è uno studente ingegnoso e determinato e sarà il primo Vulcaniano ad essere ammesso a bordo dell'Accademia della Flotta Stellare. Kirk e Spock non potrebbero essere più diversi fra loro. Ma mentre lottano per trovare la propria identità e il proprio posto nel mondo, diventano due cadetti modello e molto competitivi. Kirk, guidato da una bruciante passione e Spock, dominato invece da una logica rigorosa, saranno entrambi ammessi a bordo della più avanzata nave stellare mai creata prima, la U.S.S. Enterprise, e diventeranno due arditi e temerari avversari. L'equipaggio è guidato dal Capitano Christopher Pike (Bruce Greenwood), a cui si uniscono il medico di bordo Leonard "Bones" McCoy (Karl Urban); l'ingegnere capo della nave, Montgomery "Scotty" Scott (Simon Pegg); l'ufficiale responsabile delle comunicazioni Uhura (Zoë Saldana); l'esperto timoniere Sulu (John Cho); e il diciassettenne 'enfant prodige' Chekov (Anton Yelchin).
